Immunohistochemical study of bone morphogenetic protein and cartilage matrix protein in the salivary glands, and pleomorphic adenoma


Abstract
In order to elucidate which heterogeneous expression is relevant to the pleomorphic feature of salivary pleiotropic adenoma, we explored the distribution of bone morphogenetic protein(BMP) and cartilage matrix protein(CMP) in the fetal salivary
glands(n=100), adult salivary glands(m=20) and salivary pleomorphic adenoma(n=20). BMP was slightly positive at the terminal ducts of the late early deveolpmental stage(late EDS, 16-18 weeks) and condensely positive at the apical cytoplasm of
some
acinar cells and intercalated duct cells during the early intermediate deveolpmental stage(EIDS: 19-24 weeks of gestation) and the late intermediate develpmental stage(LIDS, 25-32 weeks). BMP positivity was gradually reduced in the LDS, and the
adult
salivary glands showed weak positivity for BMP in the intercalated duct cells. Whereas CMP was strongly positive at the apical cytoplasm of acinar cells and intercalated duct cells from EIDS. And in the later fetal stage some acinar cells showed
strong
positivity of CMP.
Meanwhile the adult salivary glands showed slight positivity for CMP for in the intercalated duct cells but almost negative in the acinar cells and striated and excretory duct cells. In the pleomorphic adenoma the modified myoepithelial cells in
the
myxochondroid tissue were strongly positive for BMP and CMP.
From the above results, we presumed that the predominant expression of BMP and CMP in the intercalated duct cells of fetal salivary gland is related to the pleomorphic features of osteochondroidal tissue in the salivary pleomorphic adenoma, and
it
is
also presumed that BMP and CMP may play a role for the induction of cytodifferentiation of salivary acinar cells during the fetal period.
